A continental perspective on the timing of environmental change during the last glacial stage in Australia

The timing and duration of the coldest period in the last glacial stage, often referred to as the last glacial maximum (LGM), has been observed to vary spatially and temporally. In Australia, this period is characterised by colder, and in some places more arid, climates than today. We applied Monte-Carlo change point analysis to all available continuous proxy records covering this period, primarily pollen records, from across Australia (n = 37) to assess this change. We find a significant change point occurred (within uncertainty) at 28.6 ± 2.8 ka in 25 records. We interpret this change as a shift to cooler climates, associated with a widespread decline in biological productivity. An additional change point occurred at 17.7 ± 2.2 ka in 24 records. We interpret this change as a shift towards warmer climates, associated with increased biological productivity. We broadly characterise the period between 28.6 (± 2.8) – 17.7 (± 2.2) ka as an extended period of maximum cooling, with low productivity vegetation that may have occurred as a combined response to reduced temperatures, lower moisture availability and atmospheric CO2. These results have implications for how the spatial and temporal coherence of climate change, in this case during the LGM, can be best interrogated and interpreted.
A soil Chronosequence on Lake Mega-Frome Beach Ridges and its Implications for Late Quaternary Pedogenesis and Paleoenvironmental Conditions in the Drylands of Southern Australia

The terminal lake systems of central Australia are key sites for the reconstruction of late Quaternary paleoenvironments. Paleoshoreline deposits around these lakes reflect repeated lake filling episodes and such landforms have enabled the establishment of a luminescence-based chronology for filling events in previous studies. Here we present a detailed documentation of the morphology and chemistry of soils developed in four well-preserved beach ridges of late Pleistocene and mid-to-late Holocene age at Lake Callabonna to assess changes in dominant pedogenic processes. All soil profiles contain evidence for the incorporation of eolian-derived material, likely via the formation of desert pavements and vesicular horizons, and limited illuviation due to generally shallow wetting fronts. Even though soil properties in the four studied profiles also provide examples of parent material influence or site-specific processes related to the geomorphic setting, there is an overall trend of increasing enrichment of eolian-derived material since at least ~ 33 ka. Compared to the Holocene profiles, the derived average accumulation rates for the late Pleistocene profiles are significantly lower and may suggest that soils record important regional changes in paleoenvironments and dust dynamics related to shifts in the Southern Hemisphere westerlies.

Automated Three-Dimensional Tracing of Neurons in Confocal and Brightfield Images

Automated three-dimensional (3-D) image analysis methods are presented for tracing of dye-injected neurons imaged by fluorescence confocal microscopy and HRP-stained neurons imaged by transmitted-light brightfield microscopy. An improved algorithm for adaptive 3-D skeletonization of noisy images enables the tracing. This algorithm operates by performing connectivity testing over large N × N × N voxel neighborhoods exploiting the sparseness of the structures of interest, robust surface detection that improves upon classical vacant neighbor schemes, improved handling of process ends or tips based on shape collapse prevention, and thickness-adaptive thinning. The confocal image stacks were skeletonized directly. The brightfield stacks required 3-D deconvolution. The results of skeletonization were analyzed to extract a graph representation. Topological and metric analyses can be carried out using this representation. A semiautomatic method was developed for reconnection of dendritic fragments that are disconnected due to insufficient dye penetration, an imaging deficiency, or skeletonization errors.
